Yes, the debit card will be blocked instantly via IB/MB/Saksham etc. Once blocked you will not be able to use your debit card for any transaction.
You will receive a confirmation message once your debit card gets blocked. Another message will be sent on issuance of your new debit card along with the details if you choose Block & Replace option.
